CryoEM FROM CELLS TO MOLECULES: Biology and Dynamics at the Mesoscale

In the last 3-4 years, thanks to several new technological developments, CryoEM has gone from producing structures at 5-25 Å resolution to now routinely producing structures at 2-4 Å resolution. Since CryoEM images single molecules, this technique directly challenges the X-ray crystallography as the go-to method for macromolecular complexes. Furthermore, for these mesoscale objects, dynamics also plays a critical role, in ways we are only beginning to understand. The same technology which has permitted such high resolutions also permits more detailed structural studies of cells. In this talk I’ll describe electron microscopy from a (once) physicist’s perspective, and describe via specific examples how this technology is leading us to new insights into cellular d macromolecular function.
